问题一：LN-229细胞；人脑髓母细胞瘤细胞长得很快很好，30万一瓶，3天就长满，也没出现污染，死细胞也较少。但是消化细胞后，用培养基中和消化液，在细胞计数板上可以看见细胞粘成一小团团的，无法准确计数，很影响种板的准确性，但传代后细胞铺板均匀，生长得也很好。我尝试了很多次细胞计数，每次都出现细胞成团的问题。

问题解答1：遇到这种情况，办法有两个，1.就是不能等到LN-229细胞；人脑髓母细胞瘤细胞长得很满时才去消化计数，可以在密度有90%时就去计数，如果怕细胞量不够铺板可以两瓶甚至三瓶合在一起。2.如果聚团还是很严重，我就把一个细胞团默认为3-4个细胞这样来计数，发现这样计数跟实际细胞数也没有太大出入，之后我就一直这样做了。我查了很多资料，都说这个细胞容易抱团生长，因为有突触，所以容易连在一起，为了避免过度消化以及吹打。

问题解答2：针对你说的抱团情况，我的处理方法，就是用PBS轻轻吹洗细胞，目的是把贴壁不好的细胞弃掉，你这样重复几次后LN-229细胞；人脑髓母细胞瘤细胞状态就能恢复了，还是强调一下，就是消化是不能消化过度，吹打时也要轻柔。另外血清浓度不要太高，这样容易导致细胞加速老化，一般10%就可以了。1562    NGS     神经元生长因子    Neuronal Growth Supplement    5 ml
